A mutant mouse line (Mutant-X) is discovered that is defective in generating IgG or IgA antibody responses to immunization with the inactivated influenza A virus vaccine. As a first step in determining the gene responsible for the immunodeficiency, T lymphocytes and B lymphocytes are separately isolated from wild-type or Mutant-X mice and mixtures of cells are transferred into Rag-deficient recipients, which are then immunized with the Influenza A vaccine as shown in Figure.
